Best practice guidelines for molecular analysis of colorectal polyposis: familial adenomatous polyposis coli (FAP) and MUTYH-associated polyposis (MAP)

چکیده
Background:. UK Clinical Molecular Genetics Society (CMGS) consensus best practice guidelines for molecular analysis of familial adenomatous polyposis coli (FAP) were published in 2000. Technological developments in molecular testing for FAP together with the clinical and molecular characterisation of MUTYH-associated polyposis (MAP) led to the need to update the original FAP guidelines which were “retired” in December 2007. This update presents consensus best practice in the molecular analysis of clinically related colorectal polyposis syndromes of FAP and MAP. Methods: Testing and reporting guidelines have been drawn up and agreed in accordance with the procedures of the UK Clinical Molecular Genetics Society. Results: A practical set of molecular genetic testing and reporting guidelines has been developed for both familial adenomatous polyposis coli and MUTYH-associated polyposis. In addition, advice is given on appropriate reporting policies, including advice on test sensitivity and recurrence risks. Conclusion: An agreed set of practice guidelines has been developed for the diagnostic molecular genetic testing of colorectal polyposis.

Familial adenomatous polyposis (FAP) is a hereditary autosomal dominant cancer syndrome, results from germ line mutation or deletion of the Adenomatous Polyposis Coli (APC) gene on chromosome 5q21. Patients with FAP suffer from multiple polyps mainly at the colorectal region as well as other parts of the gastrointestinal tract, which has propensity to transform into carcinoma. FAP has also...

Familial adenomatous polyposis (FAP) is an inherited disorder that represents the most common gastrointestinal polyposis syndrome. Germline mutations in the APC gene were initially identified as responsible for FAP, and later, several studies have also implicated the MUTYH gene as responsible for this disease, usually referred to as MUTYH-associated polyposis (MAP). FAP and MAP are characterize...

MAP (MUTYH-associated polyposis) is a syndrome, described in 2002, which is associated with colorectal adenomas, with enhanced colorectal carcinogenesis. This review synthesizes the available literature on MAP and outlines its pathogenesis, association with colorectal tumorigenesis, screening, treatment, and the subtle differences between it and its close cousins-FAP and AFAP. The preponderance...

Familial adenomatous polyposis is characterized by over 100 colorectal adenomas in the colorectum. The disease equally affects both sexes, with an incidence estimated at 1.14025-1.8300. The disease is premature in people with familial adenomatous polyposis. Patients suffering from familial adenomatous polyposis have a range of extra-intestinal diseases such as papillae, gastric, small intestine...
